摘要:脂肪酶是一种高效、专一的生物催化剂。但是游离状态的脂肪酶不易被回收重复利用,因此由于其价格的高昂,从而大大限制了脂肪酶的大规模应用。通过研究发现,将酶固定在温敏性水凝胶上可大大提高酶的重复使用率。温敏性水凝胶由于它的温敏性特征而引起了人们的广泛兴趣。目前,合成温敏性水凝胶的单体主要是N-异丙基丙烯酰胺(NIPAM),本文将分别采用PHEMA和PAEMA作为线型大分子贯穿于NIPAM形成的聚合物网络中,制备NIPAM/PHEMA和NIPAM/PAEMA两种半互穿网络水凝胶。通过测定固化酶的酶活、活力回收率、操作稳定性、热稳定性以及凝胶的溶胀-去溶胀等性能来比较NIPAM/PHEMA和NIPAM/PHAMA两种半互穿网络水凝胶在脂肪酶固定化方面上的优劣。60113

Title  The Immobilization of Enzyme on Thiol Fixed Hydrogels System

Research

Abstract

Lipase is a kind of efficient and specific biological catalysts. But the free state of lipase is not easy to be recycled use again, So because of its high price, the large-scale applications of lipase are greatly limited. Through the study, researchers found that fixing enzyme on the thermosensitive hydrogels can improve the repeat utilization of enzymes. Because the lipase is likely to be fixed in big pore aperture, this not only increased the carrier content of the unit load, but also increased the activity of unit enzyme. This article is going to achieve the purpose by taking hydrogel as the carriers and fixing lipase with the glutaraldehyde. Temperature-sensitive hydrogel has attracted intensive interest because of its thermosensitivity. At the present most of the monomers that are used to polymerize the hydrogel are N-isopropylacrylamide(NIPAM). In this article, we are going to prepare the NIPAM/PHEMA half interpenetrating network hydrogel and on NIPAM/PHAMA half interpenetrating network hydrogel by using PHEMA and PAEMA as linear macromolecules to cross through the polymer network formed by NIPAM. We have compared the performance of the immobilization of lipase on NIPAM/PHEMA half interpenetrating network hydrogel and on NIPAM/PHAMA half interpenetrating network hydrogel by determining enzyme activity, activity recovery, operation stability, thermal stability of the immobilized enzyme and swelling and deswelling properties of the hydrogels.
