现已有大量文献报道关于某些金属离子可以和一些错配碱基形成特殊的稳定结构的研究。自然碱基对与这种由金属离子介导形成的碱基对的区别在于,前者通过氢键结合以形成稳定的结构,后者通过金属离子来稳定结构。目前比较常用到的金属介导的碱基对特殊结构有胞嘧啶(C)与金属离子(Ag+)形成的C-Ag+-C结构和胸腺嘧啶(T)与金属离子(Hg2+)形成的T-Hg2+-T特异性结构。本文利用Hg2+介导T碱基配对形成的T-Hg2+-T特异性结构。该结构的形成不仅可以使两条富含T碱基的DNA单链形成稳定的双链结构,而且还可以使单链DNA的结构发生变化,使其折叠成双链。DNA和金属离子的这种特殊性质为传感器的研究提供了一种很好的思路。
