摘要:油菜与模式植物拟南芥都属十字花科,两者基因组的编码序列保守性较强,拟南芥中很多功能和结构基因在油菜中有相似或相同的功能。大量的比较基因组研究报告了油菜与拟南芥和其他十字花科植物拥有众多共线性区段,说明了两者在基因组结构上具有保守性和基因之间的共线性,使得人们能够通过拟南芥的生物信息和DNA资源库用于对油菜基因组的研究,从而加快油菜作物的遗传改良。我们利用拟南芥中一个C2H2锌指蛋白At3g23140,通过NCBI中的protein-BLAST获取与它同源的油菜C2H2中的蛋白序列,并且了解其结构特点和基本信息,分析其蛋白序列差异和进化事件,并预测了油菜中同源的C2H2锌指蛋白的高级结构和功能,表明油菜与拟南芥同源的C2H2的锌指蛋白在生长发育中具有相似的功能。研究表明,油菜C2H2型锌指蛋白共8条序列,长度不等,最长175AA,最短135AA,等电点在6.12-9.15之间,整体作用环境偏中性到碱性。油菜锌指蛋白序列均含有1个锌指结构区段,定位于细胞质或细胞外。大多数序列的二级结构以螺旋和无规则卷曲为主。29224

Mining and appraisal of rapeseed C2H2 zinc finger protein
Abstract: Canola and model plant Arabidopsis are both belonging to cruciferae plant, their coding sequence of the genome are very conservative, a lot of function and structure gene in Arabidopsis thaliana have similar or same functions in rapeseed. A large number of comparative genome research reported that Brassica napus, Arabidopsis and other cruciferae plants have many colinearity sequence in genomic, they are conservative on genome structure and gene composition, which enables people use the information and gene function Arabidopsis biological to research their homology gene information and function in rapeseed genome, to accelerate and promote genetic improvement for rapeseed crop. In the paper, a C2H2 zinc finger proteins in arabidopsis thaliana (At3g23140) as template sequence, a protein-blast was conducted in the NCBI databank to find its homologous from rapeseed protein sequence databank, then, the structure characteristics and basic information, evolutionary information of homologous et al were obtained using bioinformatics analysis method. At last, the homologous and as template sequence (At3g23140) were compared and analyzed to predict the protein senior structure and function, which indicated that the function of C2H2 zinc finger protein in Rapeseed is similar to in Arabidopsis in the growth and development. the results showed that the C2H2 zinc figure protein in Brassinca napus including 8 sequences, their length are different with a range from 135AA-175AA and with a different PI from 6.12-9.15, so they act in alkali enviorment. They were located in extracellular or cytoplasmic. Their secondary structure main include helix and random coil.
