摘要:本试验旨在研究IGF2基因3072位点多态性对猪肌内脂肪含量的影响。试验对103头苏淮猪统一屠宰,并采集了苏淮猪耳样和胸腰结合处背最长肌样品。将耳样浸入70%酒精溶液管并置于-20℃冰箱保存,采集背最长肌样品后立即抽真空包并放入-20℃冰箱冷冻保存。利用酚/氯仿法提取耳样DNA,利用索氏抽提法提取背最长肌中肌内脂肪并测定脂肪含量。设计IGF2基因第3内含子3072A>G位点引物并运行PCR,将PCR产物送与公司测序。最后将苏淮猪背最长肌肌内脂肪含量与基因型进行关联性分析。结果发现:本试验的103头苏淮猪肌内脂肪含量(IMF)的平均值是2.03%±0.60,范围是1.02%-4.08%,IGF2基因3072位点是A>G突变 ,试验发现AA型个体有17头,其肌内脂肪平均值是2.17%±0.66;AG型个体有44头,其肌内脂肪平均值是2.30%±0.58;GG型个体有42头,其肌内脂肪平均值是2.18%±0.61。通过基因型和肌内脂肪的表型数据关联性分析发现:AA型个体与AG型个体的肌内脂肪含量没有显著性差异(P>0.05),AA型个体与GG型个体的肌内脂肪含量没有显著性差异(P>0.05),GG型个体与AG型个体的肌内脂肪含量没有显著性差异(P>0.05)。由此可见,苏淮猪肌内脂肪含量与IGF2基因3072位点的多态性与苏淮猪群体肌内脂肪含量的差异没有显著性相关(P>0.05)。29702

The effects of IGF2 gene polymorphism on intramuscular fat content in Suhuai Pigs
Abstract:The aim of this study was to study the effect of polymorphism of IGF gene on intramuscular fat content in pigs. A total of 103 suhuai pigs were slaughtered and the specimens of ear ,dorsi muscle of suhuai pigs and thorax were collected. Soak ear sample into 70 % alcohol solution tube and stored in freezer at - 20 ℃, and collect the sample of the longest muscle and then take it into the vacuum bag and then put it in - 20 ℃ refrigerator. The ear DNA was extracted by phenol / chloroform method, and the intramuscular fat was extracted and the fat content was determined by soxhlet extraction. The prime of IGF2 gene’s third introns 3072A>G locus was designed and PCR was sent to the company to sequence. Finally, the correlation between intramuscular fat content and the genotype of suhuai pigs was analyzed. The results showed that the average intramuscular fat content ( IMF ) of 103 pigs was 2.03 %± 0.60, the range was 1.02 % -4.08 % and the 3072 locus of IGF2 gene was A > G mutation . the experiment finds that the AA phenotype inpidual was 17, its intramuscular fat average is2.17%±0.66; The AG phenotype inpiduals was 44, and the average intramuscular fat was2.30%±0.58. The GG phenotype inpiduals was 42, and the average intramuscular fat was2.18%±0.61.Through the genotype and the associational analysis of phenotype of intramuscular fat, there was no significant difference in intramuscular fat content between AA and AG inpiduals( p > 0.05 ), there was no significant difference in intramuscular fat content between AA and GG inpiduals ( p > 0.05 ), and so like GG and AG inpiduals. AS a result ,there was no significant correlation between the intramuscular fat content and the 3072 locus polymorphism of IGF gene in suhuai pigs ( p > 0.05).
