摘 要:本研究以籼稻C418为试材,通过无菌苗培养、外植体的筛选和激素组合配比,进而构建籼稻的体胚诱导体系。试验以籼稻根为外植体,置于含有不同浓度NAA和2,4-D的MS培养基上,在黑暗处理下进行愈伤组织和体胚诱导。将体胚置于分别含有TDZ与BAP的MS培养基中进行光照诱导胚萌成苗。结果表明:黑暗条件下,在MS+10mg/L NAA与MS+10mg/L 2,4-D的复合组合对籼稻体胚诱导效果最佳,而MS+5mg/L NAA与MS+5mg/L 2,4-D的激素配比和MS+15mg/L NAA与MS+5mg/L 2,4-D的组合效果次之。MS+5.0mg/L BAP+0.1mg/L GA3组合对体胚催萌效果最佳。通过分析不同发育时期体胚的形态学和组织学试验结果,最终成功构建了籼稻稳定、高效的类蛙卵胚体诱导体系。31485

The Construction of Frog Egg-like Body Induction System of Indica Rice
Abstract: The indica rice C418 was taken as experimental material in the study, the somatic embryogenesis induction system of indica rice were established by aseptic seedlings culturation, different explants screening and different plant growth regulators combination. The root of indica rice was made as the basic explant, on MS medium supplemented with 2,4-D or NAA with different concentration under darkness, for inducing the formation of callus and somatic embryos. The somatic embryos were cultivated on MS medium supplemented with BAP or TDZ respectively for plant regeneration. The results showed that: MS+10mg/L NAA+10mg/L 2,4-D was the optimum combination for the embryos induction, and the effect of the combination MS+5mg/L NAA+5mg/L 2,4-D and the combination MS+15mg/L NAA+15mg/L 2,4-D was not better than above combination. The combination MS+5.0mg/L BAP+0.1mg/L GA3 with the best regeneration rate on all three kinds of explants. The morphological and histological analyses of somatic embryos at different development stages revealed that the stable and high-efficiency frog egg-like body(FELB) of indica rice was established.

籼稻蛙卵胚诱导发生体系研究
引言
籼稻,是栽培稻的一个亚种,是最先经由野生稻驯化而产生的一种栽培稻。籼稻株高一般在1m以上,稻秆较软,叶片较宽,色泽呈现淡绿色,株型的松散程度适中,剑叶开度较小。叶片上的茸毛较多,而且有短小茸毛散生在颖壳之上。穗长大约20cm,每穗能结粒120左右,结实率可以达到80%以上,而且成穗率也在80%以上,平均的干粒重能达到25.6g,但是谷粒狭长且容易脱落,颖尖透明,无芒或芒尖较短。米粒中的淀粉含量很高,直链淀粉含量能达到16%左右,精米率能达70%左右,垩白粒率低于30%,蛋白质含量可达8%,赖氨酸含量可达30%。与另一种栽培稻粳稻比较:籼稻的分蘖能力较强;叶色淡绿,叶片比较宽,叶面上的茸毛比较多。稃毛短且少,谷粒细长,成熟时容易脱落,出米率稍低于粳稻。米饭黏性不强,而且膨胀性比较大;能够耐强光和耐高温,但是它的耐寒性比较差。 籼稻蛙卵胚诱导发生体系研究:http://www.751com.cn/shengwu/lunwen_27666.html
