摘要:绿色荧光蛋白(green fluorescent protein,GFP)是一种能在紫外线照射下发出强烈的绿色荧光的生物发光蛋白,存在于水母、珊瑚和水螅等腔肠动物体内。自2008年10月8日,美籍日裔科学家下村修(Osamu Shimomura)、美国科学家马丁•沙尔菲(Martin Chalfie)和美国华裔科学家钱永健(Roger Y.Tsien)三人因为发现和改造绿色荧光蛋白而获得了当年的诺贝尔化学奖以来,GFP凭借其具有稳定的荧光和构建载体方便等性质常被用来作为细胞表面展示系统的标记物,用来展示不同种类的外源蛋白,逐渐被探索应用于各种生物技术领域。本研究通过以冰晶核蛋白inaQ-N片段为载体蛋白,GFP为乘客蛋白,利用了质粒提取、引物设计、PCR扩增、凝胶电泳、DNA体外重组等技术,构建了展示GFP的大肠杆菌细胞表面展示体系,同时,对重组菌的荧光性质进行了检测,结果显示荧光效果达到预期目标。8839

The construction of cell surface display system using green fluorescent protein(GFP)
Abstract:Green fluorescent protein (GFP) is a bioluminescent protein which can issue a strong green fluorescence under UV irradiation, and present in the coelenterates’ body, such as jellyfish, coral and hydra. Since October 8, 2008, the three man, Osamu Shimomura、Martin Chalfie and Roger Y.Tsien won that year's Nobel Prize in chemistry for the discovery and transformation of the protein, GFP is often used as marker of cell surface display system to display different types of exogenous proteins by virtue of its stable fluorescent and easy to build carrier, which gradually being explored in various field of biotechnology. In this study, a cell surface display system in E. coli was constructed using the N-domain of the ice protein inaQ -N as the carrier protein and GFP as the passengers . In this study, the process of plasmid extraction, primer design, PCR amplification, gel electrophoresis and DNA reconstitution in vitro were used to achieve the recombinant bacterium. In addition, the fluorescence properties of the recombinant bacterium were evaluated. The result showed that the fluorescent effect was good.
