摘 要:布罗莫结构域(BRD)是乙酰化模体的特异性互作结构域,广泛存在于各种核蛋白如HAT、ATP依赖的染色质重建复合体、解旋酶等中。人类基因组中含61个BRD基因,其突变与多种疾病的发生有关。然而,重要作物如玉米中的BRD基因及其功能还不明确。本研究采用生物信息学方法鉴定出玉米基因组中16个BRD基因。系统发育分析表明,该家族蛋白可分为2个亚族,说明其成员已经发生了分化;在系统发育树的末端发现了7对旁系同源基因,说明该家族以物种特异的方式进行了扩张。基于EST和RNAseq数据的表达模式分析表明,多数玉米BRD基因在各组织器官中均有表达,提示该家族基因可能在植物生长发育中发挥重要功能。65732

Abstract:Bromodomain (BRD) is a specific interaction domain of acetylation motif, which is widely distributed in various nuclear proteins such as HAT, ATP dependent chromatin remodeling complexes and helicases. There are 61 BRD genes in the human genome, whose mutation is tightly related to various human diseases. However, BRD genes and their functions in important crops such as maize remain unclear. In this study, we identified 16 BRD genes from maize genome by bioinformatics methods. Phylogenetic analysis showed that BRD proteins could be pided into two subfamilies, indicating that pergence has occurred among this family. Seven pairs of paralogs were found in the terminal branches of the phylogenetic tree, suggesting that the family has expanded in a species-specific manner. Gene expression analysis using EST and RNAseq data revealed that most maize BRD genes were expressed in various tissues and organs, suggesting that they may play important roles in plant growth and development.

1 引言
BRD(Bromodomain)结构域因为发现于果蝇brahma基因的表达产物而得名,与转录活化相关基因有着相似的序列。此结构域广泛存在于多种与染色质相关的蛋白及几乎所有组蛋白乙酰转移酶中[1]。BRD结构域是识别组蛋白N端尾部乙酰化赖氨酸残基的蛋白质结构域,通过介导蛋白质-蛋白质相互作用,募集重要的功能复合体。这一识别过程往往是蛋白质-组蛋白及染色质重塑的前提条件。BRD结构域由110个氨基酸组成[2],其结构域折叠方式具有独特且高度保守的特点:采取一种全α折叠的形式,由四个α螺旋聚合形成[3]。
