摘 要:本实验是在安琪甜酒曲中提取酵母,配制YPD培养基采用梯度稀释接入培养基进行活化,条件为30℃,24 h。接着配制YPD培养基采用梯度稀释进行初选,条件30℃培养24 h。采用点样法进行复选,条件30℃培养24 h。最后采用划线培养进行酵母显微镜检测。将酵母接种到液体YPD培养基进行扩大培养,条件28℃、150 r/min、培养24 h。采用测5组数据求平均值法测生长曲线,每隔2 h用分光光度计在600 nm下测一次吸光光密度(OD)值,测13组数据,作图分析。最后破碎酵母以便提取所需蛋白,利用柱层析进行初步分离酵母Pim1蛋白酶和二羟酸脱水酶,再利用聚丙烯酰胺凝胶电泳进行检测和分析。再次进行柱层析纯化酵母Pim1蛋白酶和二羟酸脱水酶,用聚丙烯酰胺凝胶电泳进行蛋白质降解的检测。9418

Yeast Pim1 protease to the degradation of dibasic acid dehydration
Abstract:This experiment isolated yeast strains from Anqi wine Jiuqu. They were incubated with gradient dilution using potato medium at 30℃for 24 hours. After that, the yeast was originally screened with the medium of YPD and screened twice with spotting sample. After checked and picked under microscope, the yeast strains were transferred into YPD medium and incubated at 30℃ for 24 hours at the rotating speed of 150 r/min, and the growth curve of yeast was determined. Finally, the yeast strain organisms were centrifuged for cell crush and the extraction of Yeast Pim1 protease and dibasic acid dehydration. The Pim1 proteases were primarily purified with protein separation column chromatography, and detected by polyacryamide gel electropHoresis.To conduct another column chromatography purification yeast Pim1 protease enzyme and dibasic acid dehydration. Using polyacrylamide gel electrophoresis detection of protein degradation.

引言
