摘要以蝉花粉末作为原料,采用苯酚-硫酸法,并利用标准曲线建立多糖提取率的计算方法。以水作为溶剂,将提取温度、提取时间、料液比和提取次数4个单因素作为影响因素。在单因素试验优化的基础上,通过L16(45)正交试验研究4个单因素对蝉花多糖提取率的影响,从而确定蝉花多糖最佳提取工艺条件。通过实验表明,提取温度对于蝉花多糖提取率的影响是最主要的因素,而提取时间对于其影响相对较小。当提取温度90℃、料液比1:40(g:L)、提取时间60min、提取次数3次时,蝉花多糖的提取率最大。在这个最佳提取条件下,蝉花多糖的提取率为7.23%。61312
Abstract Cordyceps sobolifera powder as raw material,using phenol-sulfuric acid method,and the establishment of polysaccharide extraction rates were calculated by using the standard curve.With water as extraction solvent,extraction temperature,extraction time,solid-liquid ratio and extraction times of four single factors as influencing factors. On the basis of the optimization of single factor experiment,through L16(45) of four single factor orthogonal test to study on the extraction rate of polysaccharides in Cordyceps sobolifera,so as to determine the optimum extraction conditions of Cordyceps sobolifera polysaccharide. The experiments showed that the extraction temperature was the main factor for Cordyceps sobolifera polysaccharide extraction rate,but extraction time was relatively small. When the extraction temperature of 90℃,solid-liquid ratio of 1:40(g:ml),extraction time of 60min and extraction 3 times,the extraction rate of polysaccharide of Cordyceps sobolifera was the largest. Under this optimal extraction condition,the extraction rate of Cordyceps sobolifera polysaccharide was 7.23%.
