    摘要:真核生物中存在广泛的可变剪切现象,可以使一个 mRNA前体产生不同成熟 mRNA,从而使一个基因产生多个转录本和多个蛋白产物,提高了基因功能多样性。SR 蛋白是剪切过程的重要参与者。拟南芥中有 18 个 SR 蛋白,分为SR 亚家族、RSZ亚家族、SC 亚家族、SCL 亚家族、RS 亚家族、RS2Z 亚家族。目前有文献报道对拟南芥 SR 家族成员的突变体在强光照胁迫、镉胁迫、盐胁迫等胁迫下的研究,但冷胁迫、磷胁迫方面涉及 SR 蛋白调控的分子机制的文献不多。植物可能通过对逆境胁迫基因的可变剪切来调控逆境胁迫基因的表达来适应逆境。植物对逆境的生理适应能通过表型变化体现出来。研究拟南芥 SR 突变体表型在冷胁迫、磷胁迫下与野生型的差异具有重要的意义,能够探究哪些 SR 蛋白成员与冷胁迫、磷胁迫具有相关性。本次实验测定了 SR 突变体(T-DNA插入突变体)与野生型(Col-0)在冷胁迫下地上部分长度、根长,磷胁迫下地上部分、根部的鲜重及有效磷含量。atsr34b、atrsz21 对冷胁迫较为敏感,而且在缺磷条件下根部有效磷含量显著低于野生型,atrs31 地上部分长度在冷胁迫下显著大于野生型,在缺磷条件下地上部分鲜重、根部鲜重,地上部分有效磷含量显著大于野生型。25298

    Phenotype of SR mutants in Arabidopsis under cold stress and phosphorus stress
    Abstract:Most eukaryotic genes contain introns that are spliced from the pre-mRNAs. Alternat ive splicing
    is a kind of post-transcriptional mechanism.  In the process of alternative splicing, different  mRNAs are
    produced from a single pre-mRNA.  As a result,  different isoforms of proteins are produced.
    Serine/arginine-r ich (SR) proteins play an important role in the splicing process of pre-mRNAs.  Six
    subfamilies of SR genes are discovered in  Arabidopsis,  including SR, RSZ , SC, SCL, RS and RS2Z
    subfamily. Results from previous studies show mutation of SR gene influence on adaptation to other abiotic
    stresses in Arabidopsis such  as adjustments to light changes ,Cd tolerance and salt stress, but  the functions
    of  SR proteins on cold stress or  phosphorus stress  are largely unknown. Maybe plants have built up
    alternat ive splicing mechanisms to regulate expression of cold-induced genes to cope with stress.
    Phenotypes are an indicator of physiological adaptability to abiot ic stresses. It is of great significance to do
    research into phenotypic distinction between SR mutants and wild type under cold stress and  phosphorus
    stress. As a result, which members of SR proteins are related to cold stress and phosphorus stress can be
    figured out.  In this experiment, we do research into SR mutants(T-DNA insertion mutants)  and wild
    type(Col-0) of Arabidopsis .shoot length and root length under cold stress, fresh weight of shoots, roots and
    available phosphorous content of shoots, roots under phosphorus stress. We find atsr34b and atrsz21 are
    sensitive to cold stress, and they have  significantly lower available phosphorous  contents  in roots  than
    Col-0  under  Pi deficiency condition.  atrs31  has signif icantly  larger  shoot length  than  Col-0  under cold
    stress as well as significant ly larger fresh weight and available phosphorous content  in shoots than Col-0
    under Pi deficiency condition.
