    摘 要:本文研究建立了一种同时测定柿叶中儿茶素、橙皮素、熊果酸和齐墩果酸含量的方法并考察了该方法对其他植物的适用性。采用乙醇作为提取剂,超声波提取法对柿子叶、八角金盘叶、八角金盘果、女贞果进行提取,建立了四种标样的标准曲线并考察了它们的线性范围和重现性,并用建立的方法对提取物进行了测定及回收率的研究。采用ZORBAX Eclipse Plus C18(4.6×100mm,3.5μm)色谱柱,以甲醇-乙腈-乙酸铵(55:27:18)为流动相,采用等度洗脱,柱温为25℃,检测波长采用波长程序。儿茶素、橙皮素、熊果酸、齐墩果酸的线性范围分别为0.056~0.28μg,0.056~0.28μg,0.0344~13.76μg,0.059~2.96μg,相关系数均大于0.999,检测限分别为0.099mg/L、0.045mg/L、0.641mg/L、0.736mg/L。上述结果表明,本文建立的方法可用于柿叶等植物中黄酮和三萜类化合物的提取与同时测定。64085

    Abstract: In this paper, a method was established for the simultaneous determination of  catechin, hesperetin, ursolic and oleanolic acid in Persimmon leaf and investigate the applicability of the method to other plants.Persimmon leaves, Fatsia leaves, Fatsia fruit,Privet fruit were extracted using ethanol as the extraction agent under the ultrasonic condition, the establishment of the standard curve of four standard samples and investigate their linear range and reproducibility.These four kinds of standard sample compounds were measured with established method and some researches also done about their recoveries.Using ZORBAX Eclipse Plus C18 (4.6 × 100 mm, 3.5μm) chromatographic column with a mixture of methanol - acetonitrile - ammonium acetate (55:27:18) as the mobile phase, using isocratic elution, the column temperature was 25 ℃, the detection wavelength using wavelength programThe linear range of catechin, hesperetin, ursolic acid, oleanolic acid were 0.064 ~ 0.32μg, 0.056 ~ 0.28μg, 0.0344 ~ 13.76μg and 0.059 ~ 2.96μg, the correlation coefficients were greater than 0.999 and the detection limits were for 0.099mg / L,0.045mg / L,0.641mg / L,0.736mg / L.These results indicate that the method established in this paper can be used to separate the persimmon leaf flavonoids and triterpenoids and simultaneously.

     1  引言

    柿叶是柿树属植物柿干燥或者新鲜的叶。我国是世界上柿树主要生产国,生产产量占世界总产量一半以上,在我国柿叶资源特别的丰富,医疗保健作用确切。柿叶不仅含丰富的维生素C、黄酮甙和其他多种氨基酸等有益的成分[1],而且据研究报道柿叶黄酮具有多种药理、临床作用,例如:具有提高机体免疫功能、软化血管增强血管弹性、降低血脂、利尿通便、消肿减肥和消暑解渴安神、美容等功效,并促进机体生成抗癌、抗流行病病毒的干扰素,从生理上调整血压,防止脑动脉硬化、冠心病等,是一类具有较高开发前景的天然植物资源[2]。
