摘要:采用微波辅助法提取,利用单因素和L9(34)正交试验分析乙醇浓度、微波功率、料液比、微波时间对女贞子齐墩果酸和熊果酸提取率的影响,筛选出最优提取工艺条件,同时测定女贞子果实、叶中齐墩果酸和熊果酸的含量及其体外抗氧化活性。结果表明,女贞子中齐墩果酸与熊果酸的最佳提取工艺条件相同,为70%乙醇浓度、中火、料液比为1:25、微波3 min。此条件下,女贞子果实、叶中齐墩果酸的提取率分别为0.674 mg/g、0.873 mg/g ;女贞子果实、叶中熊果酸的提取率分别为0.512 mg/g、0.977 mg/g。女贞子提取液具有明显的抗氧化活性,其果实、叶对超氧阴离子自由基清除率分别为91.4 %、84 %,对羟自由基的清除率分别为89.7 %、55.6 %。40467

Study on the Extraction Technology of Oleanolic acid and Ursolic acid from Ligustrun Lucidum. and its Antioxidant Activities
Abstract: To explore the influence factors impacting extraction rate of oleanolic acid and ursolic acid from Ligustrun Lucidum, the optimal extraction conditions using microwave- assisted extraction were optimized by single factor and L9(34) orthogonal experiment, which included ethanol concentration, microwave power, the solid-liquid ratio, microwave-heating time and from the point of extraction rate of oleanolic acid and ursolic acid. At the same time, the oleanolic acid and ursolic acid content and antioxidant activity of fruits and stems-leaves was detected. The results showed that the optimal conditions were as follows: ethanol concentration 70%, microwave power was middle fire, the 1:25 ratio of solid-liquid, extracting for 3 min. The oleanolic acid and ursolic acid extraction rates of different parts of Ligustrun Lucidum were obtained under these conditions as follows: 0.674% of fruits, 0.873% of stem-leaf. It was also found that the extracts of Ligustrun Lucidum had different antioxidant activities. The clearance rates of superoxide anion radical in different parts of Ligustrun Lucidum were as follows: 91.4% of fruits, 84% of stem-leaf. The clearance rates of superoxide anion radical in different parts of Ligustrun Lucidum were as follows: 89.7% of fruits, 55.6% of stem-leaf.
