摘要:SEPT2蛋白广泛存在于真核生物中,为septin超家族的一员,SEPT2蛋白在胚胎及成体组织内广泛表达,其表达见于所有的脑肿瘤组织中,与肝癌、白血病、骨髓瘤、霍奇金淋巴瘤等疾病密切相关;SEPT2还参与神经递质分泌,与依靠微管蛋白形成的神经纤维有关;SEPT2通过与胞吐复合体相互作用参与胞内运输。因此对septin2蛋白的深入研究意义重大。本文采取生物信息学方法,对人septin2亚家族5条蛋白序列进行了多序列比对,并对SEPT2蛋白的基本理化性质、结构进行了分析、同时对其模建结果进行了质量评价。结果表明,人SEPT2蛋白亚家族成员,序列长度差异较小,均为不稳定蛋白,磷酸化位点多以丝氨酸磷酸化为主;二级结构β转角和扩展束较少,其中人SEPT2蛋白(gi|15680208|)的三级结构得到模建,Ramachandran 图检测结果表明此模型结构符合立体化学规则。48620

Modeling and Functional Analysis of Human SEPT2 Protein with Different Monomers
Abstract: SEPT2 protein exists widely in eukaryotic organisms, which is a member of the septin superfamily, SEPT2 protein is widely expressed in embryonic and adult tissues, it expresses in all brain tumors, liver cancer, leukemia, multiple myeloma, Hodgkin'slymphomae and so on, and it also participates in the secretion of the neurotransmitter, related to nerve fibers whose formation rely on tubulin, SEPT2 participates in intracellular transport by interacting with cell complex vomit. So it is very great significance for further studying of septin2 protein. The article adopted bioinformatic methods for the multiple sequence alignment which come from septin2 family,and analysis on physical and chemical properties as well as its structure, at the same time, in the paper, we evaluated quality of the model build.The results showed that the septin 2proteins in the human being are little differences in sequence length; besides, they are unstable proteins, and their phosphorylas sites are given priority to with serine phosphorylation, beta-turn and beta-sheet strand are less on secondary structure, and the tertiary structure of septin 2 protein was built. Finally, the Ramachandran plot showed that the modeled structure of SEPT2 protein accord with the rules of stereochemistry.
