摘要:姜黄素(Curcumin)是一种提取自姜科姜黄属植物中的有效活性成分,姜黄素在姜黄中的含量约为3%至6%。大量研究发现姜黄素具有广谱的药理活性,,尤其是它的抗肿瘤效果好、价格低廉、无毒副作用,逐渐成为研究的热点。但是,由于姜黄素存在着一些缺陷,例如在生物体内溶解性低、生物利用度差、容易被代谢排出等,因而限制了其应用。因此,研究者们为了克服姜黄素的诸多缺点,尝试过各种方法,例如进行结构改造和修饰、改变剂型,与其他物质形成包合物等等。本项目从磷化学角度出发,对具有抗癌活性的天然产物姜黄素进行酚羟基上的修饰,得到了多种姜黄素的含磷衍生物。通过我们努力所得到的姜黄素含磷衍生物较母体姜黄素而言对多种不同的人肿瘤细胞具有更好的增殖抑制作用。并且得到了具有手性的姜黄素含磷衍生物,这一合成目前尚未见报道。24572

The design and synthesis of phosphoric curcumin derivatives
Abstract: Curcumin is a kind of active principle which extracted from Plants of Zingiberaceae Curcuma Genus, and the Curcumin content was 3%~6%.A great deal of research finds that Curcumin has a broad spectrum of anticancer activity. Especially Curcumin has good anti-tumor effect, low price, and non-toxic side effects, which is becoming the research hotspot in the world. However, the greatest limitation on the application of Curcumin is some reasons like indissolubility insolubility, low bioavailability and metabolizing quickly. So people are trying in various ways to overcome many of the shortcomings of Curcumin. Such as Structural modification and retouching, Changes in dosage form or forming inclusion compounds with other
substances .etc. This project mainly embarks from the viewpoint of phosphorus chemistry, modifying the phenolic hydroxyl group on the natural product Curcumin which has a strong anticancer activity. And we obtained four kinds of phosphoric curcumin derivatives. The phosphoric curcumin derivatives have stronger inhibitory for a variety of different people than the parent substance Curcumin. And we got the first chiral phosphoric curcumin derivatives.
