    摘要:菊花(Chrysanthemum morifolium),是我国十大传统名花之一,世界四大切花之一,在市场上占有极大份额,具有极高的观赏和经济价值。栽培种植过程中,容易受蚜虫侵害,发生普遍,分布地域较广,造成严重的经济损失,独脚金内酯在抗逆中的作用逐渐得到认识,开展有关独角金内酯(SL)与菊花抗蚜虫研究对有效防治菊花蚜虫及减药栽培具有十分重要的意义。本研究以蚜虫敏感品种切花大菊 ‘神马’为研究对象,以CmCCD8基因的超表达转基因‘神马’菊花为材料,或外源独脚金内酯类似物GR24处理‘神马’,进行蚜虫虫口数量统计,“Y”型管、刺吸电位(electrical penetration graph, EPG) 等分析,明确SL对蚜虫取食行为的影响。发现:GR24处理下的虫口密度高于野生型,CmCCD8超表达转基因株系的虫口密度显著高于野生型;刺探电位图谱(EPG)显示蚜虫取食转基因株系的第一次细胞穿刺时间(Pd)、非刺吸阶段的总时间(np)和完成第一次成功刺探的时间(C)均比野生型中的时间短,且在转基因株系叶片韧皮部的取食时间显著长于野生型;GR24处理下及转基因植株对蚜虫的驱避性减弱。表明独脚金内酯具有介导菊花蚜虫敏感性的功能。26817

    The effects of strigolactone on feeding behavior of aphids on chrysanthemum
    Abstract:Chrysanthemum (Chrysanthemum morifolium), one of ten traditional Chinese famous flowers and one of four cut flowers over the world, of large market share and high ornamental and economic values. During the whole cultivated progress, aphids destroy chrysanthemum easily and universally, causes serious economic loss. The roles of strigolactone (SL) in stress tolerance in reported recently. Research of SL in resistance of chrysanthemum to aphid is essential to control aphids and non-chemical cultivation. We set up to make ‘shenma’, an aphid-sensitive variety as our research object. We used CmCCD8 over-expression ‘shenma’ lines and GR24 treated ‘shenma’ plants as material, performed aphid inoculation assay, olfactometer bioassay and electrical penetration graph(EPG) to illustrate the effects of SL on feeding behavior of chrysanthemum aphids. The results we found are as follows: GR24 treatment enhanced the aphid density compared with control plants and CmCCD8 over-expression lines are significant high. Electrical penetration graph (EPG) shows that the feeding activities of the tested aphids on the CmCCD8-OX plants differed from those on the WT plants. The time to the first cell puncturing (pd), the total duration of the non-puncturing phase (np) and the time to the first pathway phase (c) in both CmCCD8-OX plants were shorter than those in the WT plants, and the total duration of the pathway phase in CmCCD8-OX plants was longer than in WT plants significantly. GR24 treated plants and transgenic lines play negative roles in the response of chrysanthemum to aphids, which indicates SL has the function on aphids’ sensitivity.
