摘要:采用不同的方法制备羟基磷灰石,并且研究羟基磷灰石作为药物载体进行药物体外释放,对临床上治疗骨髓炎等骨科疾病具有很大的价值。
本实验采用共沉淀法先制得纳米球形碳酸钙,然后以球形碳酸钙作为模板和磷酸氢二钠在水热条件下反应,制备纳米球形羟基磷灰石。通过X射线衍射,扫描电镜,比表面积测定仪等方法对制备的纳米球形羟基磷灰石的组成、形貌和结构进行了表征。考察加表面活性剂与不加表面活性剂、反应的温度等条件对晶体的化学组成、尺寸大小、形貌和结构的影响。将制备的纳米球形羟基磷灰石置于不同浓度(分别为40mg/ml和100mg/ml)的布洛芬(IBU)溶液中,通过物理或者化学吸附作用载入药物,制备HAP/IBU缓释材料,通过紫外分光光度法对HAP/IBU的缓释材料的载药率以及体外释放规律进行了研究。23004
结果表明,在室温条件下,加入表面活性剂聚苯乙烯磺酸钠(PSS-7),Mw≈70000,以快速混合和滴加混合两种不同的方法制备的球形碳酸钙为模板,利用水热反应,能得到粒度均匀、分散良好直径约为10μm,比表面积分别为88.42m2/g和92.70m2/g的纳米球形羟基磷灰石。测定的HAP/IBU缓释材料的载药率,表明制备的HAP具备良好的载药性能,当药物浓度为100mg/ml时,其载药率分别可达143.5%和176.8%,但当药物浓度为40mg/ml时,其载药率很低,说明药物浓度对载药率有很大的影响。由药物的体外释放试验可知纳米球形羟基磷灰石能持续释药达到6天以上,表明其作为药物载体材料对药物具有很好的缓释作用。

Studies on drug delivery properties of nanometer material of hydroxyapatite
Abstract: Nanometer material of HAP, prepared with different methods, has a very high value for the clinical treatment of osteomyelitis and other bone diseases as a drug delivery system for drug release.
In the preparation of nanometer HAP, nanometer spherical calcium carbonate was firstly got in the same precipitation law system, then it was reacted with disodium hydrogen phosphate under hydrothermal conditions. The prepared nanometer spherical HAP was characterized by X-ray diffraction, scanning electron microscopy and surface area analyzer to investigate its composition, morphology and structure. Effects of temperatures, adding the surfactant or not adding the surfactant on the chemical composition, size, morphology and structure of nanometer spherical HAP crystal were investigated. The specific surface was determined by the surface area analyzer. Nanometer spherical HAP, loaded with IBU-ethanol solution(40mg/ml IBU-ethanol solution and 100mg/ml IBU-ethanol solution )by physical absorption , was detected by the ultraviolet spectrophotometer to research drug release in vitro. The loading efficiency of the nanometer spherical HAP biological material was also studied.
The result showed that nanometer spherical HAP biomaterial with two different methods uniform particle size can be prepared when the synthesis temperature is room temperature, the surfactant is PSS and its Mw=70000 and the reaction time is 3h. It has a diameter of about 10μm, specific surface area of 88.42m2/g and 92.70m2 /g respectively. The drug release property of HAP in vitro was also investigated with model drug of IBU. Nanometer spherical HAP was well dispersed and its loading efficiency were 143.5% and 176.8% respectively when the drug concentration is 100mg/ml,but when the drug concentration is 40mg/ml, its loading efficiency were very low .Nanometer spherical HAP had favorable drug release behavior in vitro as a drug delivery vehicle for it could release IBU for about 6 days.
