    摘要:大豆是中国重要的粮油作物,在农业中占有重要的地位。作物品种纯度及真实性的准确、快速鉴定是作物品种资源保存、研究和利用过程中的重要环节,因而科学准确的鉴定大豆品种、分析大豆品种纯度和种质资源的遗传变异,对于品种的审定、保护及种质材料评价等具有重要意义。本实验以Jack,大黄,猴毛和黄河四个大豆品种作为实验材料,采用SDS-PAGE电泳法获得大豆四个品种样品储藏蛋白的电泳图谱并分析其多态性;同时检测SDS-PAGE电泳法对于大豆品种纯度测定的可行性。通过对实验所得电泳图谱的观察分析,发现四种大豆品种的纯度都很高,分别为99%、99.67%、98%和99.33%;并且在检测试验中也发现四个品种间的储藏蛋白图谱差异很小,混合样品的纯度分别为98.67%、99.67%、98%和99.33%,不能只通过谱带判断品种间的差异,需要进一步精确化实验。41915

    Detection of the Authenticity and Purity of Soybean Seeds by Protein Electrophoresis

    Abstract: Soybean is an important grain and oil crops in China, occupying an important position in agriculture. The rapid and accurate identification of crop varieties’ purity and authenticity is a vital link in the process of crop variety resources’ preservation, research and utilization. Therefore, scientific and accurate identification of soybean varieties, analysis of soybean purity and the genetic variation of soybean germplasm are significant for the new species’ validation, protection and evaluation of germplasm materials. In this study, SDS-PAGE electrophoresis was used to obtain the electrophoretic patterns of soybean protein storage samples and analyzed the polymorphism of four soybean varieties of Jack, Rhubarb, Monkey Mao and Huanghe which are experimental materials. Feasibility of SDS-PAGE electrophoresis in Determination of Purity of Soybean Varieties is detected as well. Through the observation and analysis of the experimental electrophoresis patterns, it was found that the purity of the four soybean varieties was very high, for 99%, 99.67%, 98% and 99.33% respectively. In the test, it was found that the storage protein pattern of the four cultivars was very small, therefor, the purity of the mixed samples was 98.67%, 99.67%, 98% and 99.33%, the differences among the varieties need further refinement experiment rather than basing on patterns.
