摘要:本实验以乙醇为溶剂提取黄芩和槐米中的总黄酮,以单因素为基础,分别通过正交试验和响应面优化实验对黄芩和槐米总黄酮提取工艺进行研究,并通过还原力、羟基自由基的清除能力和有机自由基的清除能力比较黄芩和槐米提取液的抗氧化活性。由正交实验结果得黄芩总黄酮最佳提取工艺为:料液比1:50,提取时间2h,乙醇浓度为60%。由响应面优化实验研究的槐米总黄酮提取的最佳工艺条件为:提取温度70℃,提取时间4h,乙醇浓度70%。通过抗氧化性指标结果可得:黄芩和槐米对羟基自由基和有机自由基清除率达到50%时的提取液浓度分别为12500和5405.14mg/g、165.80和146.32mg/g;还原力A700=1时的提取液的浓度分别为2260和263.09mg/g。根据以上结果分析可得槐米的抗氧化活性优于黄芩。31047

Study on Extraction of total flavonoids and Baicalin in SopHora japonica and antioxidant activity
Abstract:The experiment with ethanol as solvent extraction of total flavonoids in Radix Scutellariae and flower buds of Sophora japonica L, using single factor based, respectively through orthogonal test and response surface optimization of the extraction technology of total flavonoids from scutellaria root and bud of Sophora japonica were studied experimental, and through reducing power, ability to scavenge hydroxyl radicals and organic by the radical scavenging ability compared Radix Scutellariae and Flos Sophorae extract antioxidant activity.The best extraction technology of total flavonoids from the experimental results was: the ratio of the material to liquid ratio of 1:50, the extraction time of 2h, the concentration of ethanol was 60%. By response surface optimization of optimal conditions for experimental study of Flos Sophorae Immaturus total flavonoids extraction: extracting temperature of 70℃, extraction time 4h, ethanol concentration of 70%. Through antioxidant indexes, the results show: Radix Scutellariae and Flos Sophorae Immaturus on hydroxyl radicals and organic free radical scavenging rate reached 50% when the concentration of extract were 12500 and 5405.14mg/g, 165.80 and 146.32mg/g; extraction liquid of reducing power A700=1 concentration respectively for 2260 and 263.09mg/g. According to the higher antioxidant activity than Scutellaria can be analysis of the above results of Sophora japonica.
