摘要:利用酵母发酵具有利用原料广、生产速率高并且可以完全适应工业化生产的特点,选用产朊假丝酵母作为研究的菌种。本课题的主要目的为通过对培养条件的优化,从摇床水平提高菌种的培养效果,以达到提高菌体生物体的含量。影响酵母菌生产出高含量葡聚糖的其中的一个非常重要的参数是产朊假丝酵母的发酵时候的条件。运用的主要方法为通过单因素实验确定单个发酵的最优条件,后运用正交实验的方法选出较优的发酵培养条件。本实验从单因素实验与正交实验的结论中得出:温度30℃,摇床转速180r/min,原YPD培养基中另外增加5g葡萄糖,培养12h的发酵培养条件。此时培养液中菌体浓度达到最大,且培养液中的葡聚糖含量达到0.0311g,较未进行优化条件前培养所得的0.025g,葡聚糖含量增加了19.6%。确定该条件为整个实验中产朊假丝酵母的最佳培养条件。52622

Optimization of culture process of Candida
Abstract:Single Cell Protein has a wide use of raw materials, high production rate and can fully adapt to the characteristics of industrial production, the choice utilis yeast strain as research. The main purpose of the project is through the optimization of culture conditions, to improve the effect of bacteria cultured from the horizontal shaker, in order to achieve content increased bacterial organisms. An important parameters affecting the yeast to produce high protein content is utilis yeast fermentation conditions. The main use of the experimental method to determine the optimum conditions for the inpidual fermentation by a single factor, after the orthogonal experiment method to elect the optimum fermentation conditions. In this study, from single-factor experiment and orthogonal experiment conclusions drawn: temperature 30 ℃, shaking speed 180r / min, former YPD medium in an additional 5g glucose, culture fermentation conditions for 12h. At this point the culture medium reaches the maximum cell concentration, and the culture medium glucan content of 0.0311g, compared with 0.025g culture conditions are not obtained before optimization carried glucan content increased by 19.6%.
